L’Unità di Neurofarmacologia ha come obiettivi lo studio del ruolo dei recettori metabotropici (mGlu) per il glutammato nella fisiologia e patologia, con particolare riferimento ai meccanismi di neurodegenerazione e neuroprotezione. Questi recettori (ne esistono otto sottotipi mGlu1 – mGlu8), sono degli ottimi bersagli per la terapia farmacologica poiché modulano sia la trasmissione sinaptica eccitatoria ed inibitoria nel sistema nervosa centrale, e i ligandi dei recettori mGlu sono, in principio, privi degli effetti avversi degli antagonisti dei recettori ionotropici per il glutammato, come sedazione, atassia ed alterazione dei processi di apprendimento e memoria. Il ruolo di questi recettori viene studiato sia in modelli in vitro che in vivo di patologie neurodegenerative quali la malattia di Parkinson, la malattia di Alzheimer, la malattia di Huntington, la Sclerosi Laterale Amiotrofica e le atassie spinocerebellari ereditarie. Lo studio del ruolo dei recettori mGlu si estende anche a patologie neurovascolari come l’ictus ischemico, patologie epilettiche come le assenze epilettiche, patologie tumorali cerebrali come i glioblastomi e i medulloblastomi, patologie con componente autistica come la malattia di Angelman e la Sindrome X Fragile e patologie psichiatriche come la schizofrenia.
